Exon

Meaning: The EGFR exon the variant falls in. The library covers the tyrosine kinase domain, exons 18 through 24.

Type

Meaning: What the variant does to the protein — Missense, Nonsense, Synonymous, or Insertion.
Context: Synonymous variants change no amino acid and serve as the screen's internal reference for a variant with no functional effect. They are also the distribution the classification cutoffs are drawn from.

AA change

Meaning: The amino acid change, in one-letter code (for example, T790M), or the protein-level description of an exon 20 insertion (for example, p.A763_Y764insFQEA).
Context: One row per amino acid change. Several distinct nucleotide edits can produce the same one, and the screen scores and classifies at the amino acid level, so those edits share a single verdict rather than repeating it a row at a time.

<TKI> AA resistance score

Meaning: How strongly cells carrying the variant survive that TKI, scored at the amino acid level and shown to two decimal places. Positive scores mean the variant is enriched under treatment — the drug is losing its hold. Negative scores mean it is depleted — the drug is still working.

<TKI> AA classification

Meaning: Variant-specific drug response (Resistant, Intermediate, or Sensitive), classified by LFC between treated and untreated pools using cutoffs defined from the empirical LFC distribution of synonymous variants.
Reading the table: This is the only coloured column — Resistant red, Intermediate green, Sensitive blue. The score beside it is left plain so the numbers can be compared without a second colour coding over them.

N/A

Meaning: No measurement, either because the variant fell outside the region that drug was screened across, or because the screen returned no score the classification could rest on.
