EGFRvariants reports how each EGFR protein variant responds to a tyrosine kinase inhibitor (TKI), measured by drug response and cell viability in non-small cell lung cancer (NSCLC) cells carrying an activating EGFR L858R or exon 19 deletion mutation. Variants were installed by prime editing with a custom EGFR pegRNA library, and the cells were treated with afatinib, dacomitinib, or osimertinib.
Two studies are covered — Kim et al. Nature Biotechnology (2025) and Lee et al. npj Precision Oncology (2026) — and each screened its own activating contexts against its own set of drugs.
Results are chosen in three steps, because each screen covers its own activating context and its own set of drugs. Choose a Dataset, then an Activating context, then a TKI, and press Show Results.
1. Dataset
2. Activating context — the EGFR mutation the screened cells already carry, which is what a TKI is being given for in the first place. A variant's effect is read on that background, so the same variant can score differently between contexts.
The contexts on offer are the ones that dataset screened, so the control offers a different pair under each.
3. TKI — one of the drugs screened in that context, or All TKIs to place every drug's columns side by side in one table and compare them row by row.
Under Lee et al. (2026) / Exon 19 deletion, afatinib and osimertinib were screened across exon 20 only; dacomitinib covers exons 18–24. Rows outside exon 20 therefore read N/A in the afatinib and osimertinib columns. Filter the Exon column to 20 to see only the rows all three drugs cover.

One row per amino acid change, with a score and a classification for each TKI the context covers.
